The purpose is to investigate the tendency of bacterial growth in water subjected to degassing using a degasser and to examine whether degassing that does not lead to anaerobic conditions affects the growth of fungi.

【Results】 Degassed water: The number of bacteria temporarily increased but then decreased to a level that could not be measured. There is a possibility that they have died. Non-degassed water: The number of bacteria initially increased, then decreased, and subsequently continued to increase again. After 20 days, it reached a level that could not be measured.

This is the result of investigating the decrease in vitamin C content due to the participation and alteration of vitamin C aqueous solution over time using HPLC.

Pharmaceuticals and food ingredients contain substances that can easily oxidize or deteriorate due to oxygen. When analyzing the quantities of these components using various instruments such as HPLC (High-Performance Liquid Chromatography), it poses a significant obstacle to recognizing their original values. Furthermore, in the case of pharmaceuticals and food, it is not only the decrease in potency (reduction of active ingredients) that is a concern, but there are also cases where oxidation products and deteriorated substances may have adverse effects.

Technical Report: The Effect of Degassing in Fluorescent Detection

The chromatogram shows the separation and detection of anthracene and pyrene by HPLC (high-performance liquid chromatography).

The only difference in the mobile phase used is whether it has been degassed using a degasser (R) or not; all other conditions, including sample concentration and sample volume, are exactly the same. However, even with just the difference in degassing conditions, the peak size of pyrene shows completely different results.

We have successfully achieved long-life operation of the device. The high durability of the degassing device is essential for avoiding issues with the liquid delivery pump and for flow rate reproducibility and stability.

We have developed a high-performance degassing device that supplies a solvent, free of stable dissolved oxygen and without significant vacuum fluctuations, to a delivery pump. The combination of a pulse motor and our proprietary control software, along with a self-cleaning mechanism for atmospheric introduction during device operation, has achieved a long lifespan for the vacuum pump. The permeable membrane of the degassing module uses high-performance Teflon AF (amorphous fluoropolymer), which has excellent air permeability. Therefore, even with a low volume (480 microL), our vacuum pump's performance allows for a wide range of applications from micro-delivery to semi-preparative processes. In terms of OEM support, during maintenance, the status and inspection of each point can be confirmed via an LCD display, and experiments will not be interrupted even during analysis. The effect of increased apparent density by the vacuum press is due to both vacuum and mechanical pressure. The raw material is covered on the surface of the filter drum, which is being evacuated from the inside. As the drum rotates, layers are continuously formed, leading to an increase in primary bulk density. The figure shows the relationship between the amount of air concentration that dissolves in water and the corresponding amount of oxygen concentration with respect to water temperature.

Typically, the oxygen concentration is expressed as the dissolved oxygen concentration (DO value), represented by the weight of oxygen (mg) dissolved in 1 liter of water (mg O2/L = ppm). The figure shows the concentration of dissolved oxygen as a line graph and the volume of dissolved air as a bar graph (converted for water temperature and at 0°C). The actual air volume (bubble volume) is larger because water vapor corresponding to the vapor pressure exists within the bubbles.

We compared the durability of the degassing effect when degassing online and when degassing the water in the bottle using other methods.

The degasser maintains a consistently low level of degassing for the water supplied to the flow path while allowing liquid to pass through. On the other hand, if the water in the bottle is degassed all at once, it will rapidly reabsorb atmospheric (oxygen) over time, and the degassing effect will quickly diminish.
